丹参配方颗粒HPLC指纹图谱研究

摘    要:目的:建立丹参配方颗粒的HPLC指纹图谱。方法以丹酚酸B为参照物,利用高效液相色谱梯度洗脱,测定了11批丹参配方颗粒样品;色谱柱为Waters SunFire C18(4.6mm ×250mm,5μm);流动相:甲醇(A)-0.4%的甲酸溶液(B),检测波长270nm,柱温30℃,流速1.0mL· min -1。结果通过HPLC指纹图谱分析,标示出丹参配方颗粒16个共有色谱峰,相似度均大于0.96,并确认2个已知峰为丹酚酸 B和丹参酮ⅡA。结论建立的丹参配方颗粒的HPLC指纹图谱稳定可靠,操作简便,可为丹参配方颗粒质量控制提供科学依据。

Study on the HPLC Fingerprint Chromatogram of Salvia Dispensing Gran-ule
Abstract:ABSTRACT:OBJECTIVE To establish an HPLC method for determination of the fingerprint of Salvia Dispensing Granule.METHODS 11 samples were analyzed by HPLC,with Salvianolic acid B as the references.Analysis was performed on a Waters SunFire C18 column ( 4.6mm ×250mm,5μm) with mobile phase A:Methanol and mobile phase B:0.4%Formic acid solution by gradient elution;flow rate was 1.0mL· min-1 ,λ=270nm,Column box tem-perature was 30℃.RESULTS By analyzing the Chromatogram fingerprint,16 peaks existed which included Salvi-anolic acid B and Tanshinone ⅡA.The similarity of 11 samples was more than 0.96.CONCLUSION The estab-lished HPLC fingerprint Chromatogram of Salvia Dispensing Granule methed is dependable and simple.This method can provide scientific basis for the quality control of Salvia dispensing Granule.
