雷帕霉素逆转骨髓瘤U266细胞黏附介导的耐药

摘    要:目的:探讨雷帕霉素( Rapamycin,RAPA)对骨髓瘤U266细胞的黏附作用的影响及因黏附介导的阿霉素耐药的影响。方法采用结晶紫染色法检测U266细胞与纤连蛋白( Fibronectin,FN)的黏附作用;MTT实验检测RAPA对骨髓瘤细胞因黏附介导的阿霉素耐药的影响。结果U266细胞与FN黏附1、6、12h,其黏附率分别为(35.13±4.31)%、(43.65±3.86)%和(60.38±5.27)%;用160nmol· L -1 RAPA 处理1、6、12h后黏附率分别降为(22.14±6.13)%、(21.99±5.68)%和(27.76±4.91)%,组间比较差异显著(P <0.05);当阿霉素作用时,FN黏附组细胞IC50值〔(3.16±0.29)mol· L -1〕显著高于BSA组〔(0.62±0.19)mol· L -1〕(P<0.05);FN联合RAPA组 IC50值〔(0.65±0.31)mol· L -1〕显著低于FN组(P<0.05),但与BSA联合RAPA组〔(0.58±0.22)mol· L-1〕相比无显著性差异(P>0.05),但却低于 FN组(P<0.05)。结论 RAPA能降低U266细胞与FN的黏附率和逆转黏附介导的阿霉素耐药。

Rapamycin Reverses Adriamycin Resistance Mediated by Cell Adhesion in Myeloma Cells
Abstract:OBJECTIVE To investigate the effect of RAPA on the adhesion of U266 cells to fibronectin ( FN) and cell adhesion mediated adriamycin ( Adr)-resistance.METHODS The adhesion of U266 cells to FN and drug resistance mediated by cell adhesion were determined using crystal violet staining analysis and MTT assays,respec-tively.RESULTS Crystal violet staining analysis showed RAPA could inhibit the adhesion of U266 cells to FN.When U266 cells were adhered to FN or BSA-coated wells for 1,6 and 12h,the adhesion rates were (35.13 ± 4.31)%,(43.65 ±3.86)%,and (60.38 ±5.27)%,respectively.After treatment with 160nmol· L-1 RAPA,the adhesion rates were decreased to (22.14 ±6.13)%,(21.99 ±5.68)%and (27.76 ±4.91)%,respectively ( P<0.05).When exposed to Adr,the mean IC50 value for FN-adhered cells〔(3.16 ±0.29) mol· L-1〕 was significant higher than the mean IC50 value for BSA control〔(0.62 ±0.19)mol· L-1〕 (P<0.05);the mean IC50 value for RAPA+FN group was significant lower than the FN-adhered group ( P<0.05 ) .But there was no significant dife-rence between the RAPA+FN group and RAPA+BSA group ( P>0.05 ) .CONCLUSION RAPA could reduce the adhesion rate of U266 cells to fibronectin and reverse cell adhesion mediated adriamycin-resistance.
