生长受限对胎鼠脑组织能量代谢及神经细胞凋亡率的影响

摘    要:目的:探讨宫内生长受限(FGR)仔鼠脑组织能量代谢的变化及对神经细胞凋亡的影响。方法:(1)用子宫动脉部分结扎法建立FGR模型。孕鼠分为两组:对照组10只,于妊娠17天行开腹关腹手术,术后待其自然分娩。FGR组20只,于妊娠17天行双侧子宫动脉部分结扎术,术后待其自然分娩;(2)高效液相法检测两组部分仔鼠脑组织ATP、ADP、AMP含量;(3)TUNEL法检测两组部分仔鼠脑组织细胞凋亡率。结果:(1)仔鼠脑组织ATP的含量(‰):对照组为625.01±115.32,FGR组为115.32±33.44,两组差异均有显著性(P<0.01)。ADP的含量(‰):对照组为310.76±28.91,FGR组为180.73±35.35,两组差异亦有显著性(P<0.01)。AMP含量(‰):对照组为117.36±15.12,FGR组为126.96±25.08,差异无统计学意义(P>0.05);(2)两组仔鼠大脑皮层神经细胞凋亡率:对照组为2.3±0.69个/HP,FGR组为5.05±1.17个/HP,两组差异有显著性(P<0.01);海马CA1区神经细胞凋亡率:对照组为0.53±0.26个/HP,FGR组为1.62±0.57个/HP,两组差异有统计学意义。结论:FGR仔鼠脑组织ATP、ADP含量明显减少,神经细胞凋亡率增加,脑组织能量代谢障碍可能是导致FGR仔鼠神经系统损伤的机制之一。

Abstract:Objection:To study the changes of cerebral energy metabolism in intrauterine growth restricted fetal rats and its effects on neuronal apoptosis.Methods:Pregnant rats were divided into two groups,10 in the control group,and 20 in the FGR group.FGR rat pups were created by partial occlusion of bilateral uterine artery of pregnant dams on the 17th day of gestation.ATP,ADP,AMP contents in cerebral tissues of pups were measured by high performance liquid chromatography.Neuronal apoptosis in cortex and hippocampus of newborn pups in each group were detected by TUNEL.Results:(1) The contents of ATP(‰) and ADP(‰) in control group were much more abundant than that of FGR group(625.01±115.32 vs 115.32±33.44 for ATP,P<0.01),and(310.76±28.91 vs 180.73±35.35 for ADP,P<0.01).There were no statistical difference about the contents of AMP between two groups(117.36±15.12 vs 126.96±25.08,P>0.05).(2) The ratio of neuronal apoptosis in FGR group was significantly higher than that of control group(5.05±1.17/HP vs 2.3±0.69/HP in the cortex,P<0.01),and(1.62±0.57/HP vs 0.53±0.26/HP in hippocampus CA1,P<0.01).Conclusion:The contents of ATP and ADP in cerebral tissues are significantly decreased in FGR pups,while the ratio of neuronal apoptosis is significantly higher than normal pups.Impairment of energy metabolism may be one of the mechanisms that result in nervous system injuries of FGR pups.
