rhIL-1受体拮抗剂对大鼠肾小球系膜细胞增殖和合成细胞外基质-胶原的影响

摘    要:采用肾小球系膜细胞(GMC)培养、~3H-TdR掺入及羟脯氨酸测定的方法,首次观察了重组人白细胞介素-1受体拮抗剂(rhIL-1Ra)对白细胞介素1β(IL-1β)诱导的大鼠GMC增殖和合成细胞外基质-胶原的影响。结果表明:rhIL-1Ra能拮抗hIL-1β诱导的GMC的增殖和胶原的合成;其拮抗增殖的剂量是rhIL-1β的2~50倍,拮抗胶原合成者为rhIL-1β的25~50倍。提示:rhIL-1Ra有望用于治疗由IL-1参与、介导的以系膜细胞增殖和系膜基质扩大为主要病理改变的肾小球肾炎。

Abstract:Using the methods of GMC culture,3H-TdR incorporation and measurement of hydroxyproline.we first observed the effects of rhIL-IRa on the proliferation and the synthesis of extracellular matrix-collagens induced by rhIL-1B in GMC of rats. The results showed that: 1. rhlL-lRa could antagonize the proliferation and collagens synthesis induced by rhIL-l|3. Its dosage that antagonized to the proliferation was 2~50 folds of the rhIL-lB. Its dosage that antagonized to the collagens synthesis was 25~50 folds of the rhIL-lB. It suggests that rhIL-IRa might be used to treat glomeru-lonephritis initiated by IL-1B.
