胚鼠大脑皮层神经元培养方法的改进

摘    要:目的建立一种简单、高效的胚鼠大脑皮层神经元原代培养的方法。方法17d胎鼠大脑皮质,直接机械切碎后经200目滤网研磨过滤,得到细胞悬液进行培养。结果神经元生长良好。相邻细胞的突起交错形成网络,鉴定神经元纯度为(94.7±5.6)%。结论本方法简单可行。可以获得高纯度的胚鼠大脑皮层神经元。

Abstract:Objective To establish an easier and better primary culture technique suitable to neurons of embryonic rats cortical tissure. Methods The cortex of El7 rats was isolated and minced by mechanical dissociation and filtered by strainer mesh of 200,then to conduct culture. Results The neurons grew well and typical nerver fiber net was formed. Identification the purity of neurons isolated with this method is(94.7±5.6) %. Conclusions The purity of neurons isolated with this method is high and the method is simle and a good technique.
