天花粉降糖作用有效部位的研究

摘    要:目的:观察天花粉的降糖作用并筛选其有效部位。方法:天花粉乙醇提取物用水分散后分别用石油醚、乙酸乙酯,正丁醇萃取得到3个提取物;天花粉水提物用硫酸铵沉淀,蒸馏水透析得到凝集素粗品。检测天花粉4种提取物对人肝癌细胞HepG2的葡萄糖消耗量的影响;建立四氧嘧啶糖尿病小鼠模型,对各提取物进行降血糖研究,确定天花粉降糖有效部位。结果:乙酸乙酯部位和凝集素粗品具有较强的降糖作用,其中以凝集素部位为佳。结论:凝集素为天花粉降糖的主要有效部位。为进一步开发天花粉降糖有效成分提供了理论依据。

Abstract:Objective:To study the active fraction and screen the effective parts.Method:Tested fractions were obtained by partitioning the ethanol extracts of trichosanthes kirilowi using petroleum ether、ethyl acetate and butyl alcohol.The water extracts were precipitated by ammonium sulfate and dialysised by distilled water.Human hepatocarcinoma cell HepG2 were used to assess Hypoglycemic activity of the four extracts in Trichosanthes kirilowi;Investigated the hypoglycemic activity of the extracs through the alloxan-induced diabetic mice model,and achieved the hypoglycemic parts.Result:Ethyl acetate and lectin parts exposed proper hypoglycemic activity,and lectin parts is preferred.Conclusion:Lectin parts extracted from Trichosanthes kirilowi were the main hypoglycemic parts,the study provided theory basis for further development of the effective components from Trichosanthes kirilowi.
