固定化黑曲霉细胞与固定化葡萄糖异构酶生产高含量低聚果糖

摘    要:目的 :研究固定化黑曲霉细胞与葡萄糖异构酶协同反应生产高含量低聚果糖的优化条件。方法 :以磷酸化的PVA 海藻酸钙为载体包埋黑曲霉细胞 ,与固定化的葡萄糖异构酶协同反应 ,以蔗糖为原料转化生产低聚果糖。结果 :7.5 %PVA ,1.0 %海藻酸钠包埋 10 %黑曲霉细胞 ,在 pH 6 .7的饱和硼酸 0 .75 %BaCl2 中成型 ,于 0 .7mol/LNaH2 PO4 中强化 1h。与固定化的葡萄糖异构酶一同在 5 0℃ ,pH 7.0条件下与 40 %蔗糖溶液反应 3.5h。重复反应 5批 ,平均转化率为 5 7%。结论 :用优化条件制得的固定化细胞与葡萄糖异构酶协同转化生产低聚果糖 ,重复利用性好 ,且转化率高于单独使用黑曲霉

Abstract:Purpose:The aim is to study the optimal co reaction condition of producing high content fructooligosaccharides(FOS) for immobilized cells and glucose isomerase.Methods: Aspergillus niger cells entrapped by modified phosphorylated polyvinyl alcohol(PVA) alginate gel co react with immobilized glucose isomerase to transfer sucrose to FOS.Results:10% cells entrapped by 7.5% PVA and 1.0% alginate are solidified in saturated boric acid-0.75% BaCl 2 with pH 6.7 and strengthened in 0.7 mol/L Na 2HPO 4.The immobilized cells were co reacted with immobilized glucose isomerase at 50℃,pH 7.0 for 3.5 hours.After 5 batches were repeated,the FOS average conversion rate was 57%.Conclusion:In the reaction system which is composed of the cells entrapped with optimal conditions and immobilized glucose isomerase,the re utilization ability is good and conversion rate is higher than that of the solo immobilized cell reaction.
