Abstract:Objective To investigate the effects of black rice anthocyanins(BRACs) on apoptosis and expression of caspase-1 mRNA and protein in photochemical damaged retinas.Methods Sixty Sprague-Dawley rats were randomly divided into control group(n=30) and BRACs group(n=30),and fed with basic diet with a 12 hous light-dark cycle.The BRACs group was administered with BRACs by stomach intubation at a dosage of 100 mg/kg·bw for 15 days.Meanwhile,the control group was administered with equal volume physiological saline without BRACs.Then the two groups were exposed to(3000±200) lux fluorescent light for 0-24 hous.Thereafter,the apoptosis of photoreceptors was assessed by terminal-deoxynucleoitidyl transferase mediated nick end labeling(TUNEL),and the expression of caspase-1 mRNA and protein in photochemical damaged retinas was determined by RT-PCR and immunohistochemistry,respectively.Results The apoptotic photoreceptors were increased markedly after light exposure(P0.05),and BRACs suppressed photoreceptors degeneration(P0.05).The expression level of caspase-1 mRNA and protein was increased in light damaged retinas,and BRACs down-regulated caspase-1 expression(P0.05).After light exposure,there were considerable caspase-1 positive cells,but there was a decrease in BRACs group compared to the control(P0.05).Conclusion BRACs can inhibit photoreceptor apoptosis,which maybe is related with the down-regulation of apoptotic proteins caspase-1.
