摘    要:目的建立黄酮含量测定的高效液相色谱法,并测定成都市售8种蔬菜中黄酮含量。方法采用高效液相色谱法,色谱条件:色谱柱Allsphere ODS-2Su C18柱,流动相乙腈和水(60:40),流速1ml/min,柱温为25℃,进样量10μl,检测波长320nm。结果黄酮含量在0.02~0.2mg/ml范围内呈良好的线性关系(r2=0.999),平均回收率99.3%(RSD=1.22%)。成都市售8种蔬菜中均含有黄酮类物质,芹菜和韭菜中黄酮类物质含量较高。结论建立的高效液相色谱法操作简便,分离度和灵敏度高,可适用于蔬菜中黄酮的测定。测定结果为合理的膳食指导提供了基础数据。

Determination of flavonoids contents of common vegetables in Chengdu by high performance liquid chromatography

Abstract:Objective To establish high performance liquid chromatography(HPLC) method for measuring the total contents of flavonoids,and further to detect the flavonoids contents of 8 kinds of vegetables in Chengdu city.Methods The chromatographic condition of HPLC was as follow: the chromatographic column(Allsphere ODS-2 Su C18),mobile phase(acetonitrile:water=60:40,V/V),flow rate(1.0 ml/min),and the measure wavelength(320nm),the column temperature(25℃) and injected sample volume(10 μl).Results The calibration curve of flavonoids showed a good linearity with the peak area in the range of 0.02-0.2mg/ml(r2=0.999),and the average recovery rate was 99.3%(RSD=1.22%).All the eight kinds of vegetables detected in Chengdu city contain flavonoids,and the content of flavonoids in celery and leek is higher than others.Conclusions The established HPLC method is convenient for operation,possesses better separation and higher sensitive degree,and suitable for measurement of flavonids in vegetables.Our results may provide basic datum for dietary guidance.
