凉山彝族自治州HIV流行毒株基因亚型分析

目的对16例来自凉山彝族自治州的HIV感染者进行亚型分析,监测该地区感染人群HIV毒株的流行情况。方法从16例HIV-1抗体阳性样本中分离外周血单核细胞（PBMC）,从PBMC中提取基因组DNA,直接进行巢式PCR扩增HIVenv基因c2～v5区及gag基因p17～p24区,对PCR产物进行核苷酸序列测定和分析,确定基因亚型。结果 16例样本全部为BC重组亚型,env基因的基因距离较gag基因远,gag基因在进化上与CRF07-BC更为接近。结论凉山彝族自治州的HIV流行毒株主要为CRF07-BC。

Analysis of the HIV subtypes in Liangshan Yi Autonomous region

Objective To monitor the epidemiology of HIV by analyzing HIV subtype of 16 subjects with HIV-1 in the Liangshan Yi autonomous region.Methods DNA was extracted from the PBMCs of 16 HIV-1 positive samples,HIV env（c2～v5 region） and gag（p17～p24） gene were amplified by nested-PCR using specific primer pairs and the product was sequenced directly.The acquired sequences were analyzed to determine the HIV subtype.Results All the cases were the B/C recombinant strains,the gene distance of env region is farther than that of gag region.Liangshan Yi Autonomous Region is CRF07-BC,and the prevalence of HIV-1 among the cases,phylogenetic analysis showed that gag gene of all the cases were closer to CRF07 -BC than CRF08 -BC.Conclusions The major subtype of HIV -1 among IDUs in Yi population in Liangshan is CRF07-BC.The transmition of HIV-1 tends from the IDUs to common population in this region now.
