新生大鼠大脑皮层神经细胞的体外原代培养

摘    要:本文探讨了新生大鼠出生24h内大脑皮层神经细胞原代培养的方法,建立了稳定的神经细胞培养模型。通过光镜和细胞Nissl染色、AChE染色,对培养各阶段的神经细胞进行了观察,系统地描述了它们的生长发育过程和形态特征。本方法扩大了大脑皮层神经细胞培养材料的来源。另外,对培养方法的特点进行了讨论。

Abstract:The neuronal primary culture of cerebral cortex is the one of important methods of research for morphology, function. damage,ischemia and so on of cells. People used to using embryonic neurocytes from 15-18-day old fetal rat and 68-day old fetal chicken for neuronal culture. We studied the neuronal culture methods of cerebral cortex of newborn rats (within postnatal 24h). Using the light microscopy, Nissl's staining and AChE histochemistry, we observed neurons of cerebral cortex at different culture stages. described their whole development course and morphologic properties. The results showed steady neuronal culture models, this method enlarges the material sources of neuronal culture. In addition, the characters of the culture method were discussed.
