荧光定量PCR与培养法检测泌尿生殖道支原体感染的应用

摘    要:目的 了解泌尿生殖道感染患者中支原体的感染现状。方法 以荧光定量PCR技术进行解脲支原体的检测 ;同时采用深圳浪峰生物试剂公司的支原体培养试剂盒进行了Uu的培养及药敏试验。结果  5 18份标本荧光定量PCR共检出Uu180例 ,检出率为 3 4.7% ;培养—药敏一体化试剂共检出解脲支原体 13 1例 ,阳性率 2 5 .3 % ,两法的符合率为 72 .8%。药敏结果显示 ,强力霉素、可乐必妥、交沙霉素和阿岐霉素的敏感性较高 ,分别为 5 .3 %、8.4%、4.6%和 6.1% ;而四环素、红霉素、螺旋霉素耐药率较高 ,分别为 61.8%、45 .8%和 5 1.1%。结论 荧光定量PCR技术在临床疗效的观察中具有高度的诊断价值 ,而培养法对于感染的诊断和用药的选择更有意义。Uu对强力霉素、可乐必妥、交沙霉素和阿岐霉素较敏感 ,对四环素、红霉素、螺旋霉素耐药。

Abstract:Objective To study the condition of mycoplasma infection in patients with genitourinary tract infection. Methods A FQ-PCR technique was used to assay the Ureaplasma mycoplasma; at the same time, the Uu cultivation and drug sensitive test had also been done with mycoplasma cultivation kit produced by Shenzhen Langfeng Biologic Reagent Company. Results In 518 samples, 180 cases of them were detected with Uu by FQ-PCR, the detectable rate was 34.7%; and 131 cases of them were detected with Ureaplasma mycoplasma by cultivation-drug sensitivity integral reagent, the positive rate was 25.3%, the coincidence rate of this two methods was 72.8%. The drug sensitivity showed that Doxycycline, Levofloxacin, Clarithromycin and Azithromycin had higher sensitivity; their sensitive rates were 5.3%, 8.4%, 4.6% and 6.1%, respectively. And Tetracycline, Erythromycin and Spiramycin had higher drug-resistant rate; their rates were 61.8%, 45.8% and 51.1%, respectively. Conclusions FQ-PCR technique had great diagnostic value in monitoring the clinical effectiveness, but cultivation was of great value in diagnosing infection and selecting medicine. Uu was more sensitive to Doxycycline, Levofloxacin, Clarithromycin, and Azithromycin, and it was resistant to Tetracycline, Erythromycin and Spiramycin.
