血管性痴呆大鼠模型海马乙酰胆碱和血小板激活因子含量变化的动态观察

摘    要:目的 探讨乙酰胆碱(acetylcholine,ACh)和血小板激活因子(Platelet activatingfactor,PAF)在血管性痴呆中的含量变化及意义。方法 用比色法和高效薄层层析法,分别测定血管性痴呆(vascular dementia,VaD)大鼠手术后,第2、3、5天海马ACh和PAF含量。结果 手术后第2、3、5天大鼠海马ACh含量,在模型组与对照组间均有显著性差异(P<0.05);而PAF含量两组间则均无显著性差异(P>0.05)。结论 血管性痴呆大鼠海马ACh含量的持续降低,可能是VaD发生、发展的重要原因之一;而PAF则可能主要参与急性脑缺血早期的病理生理过程,对脑缺血后期的病理生理变化意义不大。

Abstract:Objective To investigate the concentration and significance of acetylcholine(ACh) and platelet activating factor(PAF) in vascular dementia(VaD). Methods On the 2, 3d and 5d after operation , Wistar rats were killed and the concentration of ACh and PAF in rats hippocampus were measured with high performance thin-layer chromatography and colorimetric method. Results 2, 3, 5d after operation , there was significant difference in the concentration of ACh and no significant difference in PAF between the two groups. Conclusion The contin-uing decline of ACh concentration of hippocampus may be one of the reasons for VaD occurrence and development; but PAF may participate mainly in the pathophysiology course in the early stage and have little effect on the later stage of cerebral ischemia.
