RECK及MMP-2在骨肉瘤组织中的表达及意义

目的检测RECK蛋白及明胶类基质金属蛋白酶-2（MMP-2）在骨肉瘤组织表达,探讨其在骨肉瘤的发生、发展、浸润和转移中的作用。方法选择临床及病理资料完整的存档骨肉瘤标本30例,另取骨软骨瘤标本10例作为对照。采用PV-6000免疫组化方法检测RECK及MMP-2蛋白的表达。结果 RECK在骨肉瘤组织中低表达（30.0%）,在骨软骨瘤组织中高表达（80.0%）,两者相比差异有显著性（χ2=5.76,P〈0.05）;MMP-2在骨肉瘤组织中高表达（76.7%）,在骨软骨瘤组织中低表达（10.0%）,两者相比差异有显著性（χ2=11.25,P〈0.05）;骨肉瘤组织中RECK与MMP-2的表达呈负相关（r=-0.84,P〈0.05）。结论骨肉瘤组织中RECK低表达,而MMP-2高表达。RECK及MMP-2可能作为预测骨肉瘤浸润转移的重要指标。

EXPRESSION AND SIGNIFICANCE OF REVERSION-INDUCING CYSTEINE-RICH PROTEIN WITH KAZAL MOTIFS AND MATRIX METALLOPROTEINASE-2 IN OSTEOSARCOMA

Objective To detect the expressions of reversion-inducing cysteine-rich protein with Kazal motifs（RECK） and matrix metalloproteinase-2（MMP-2） in osteosarcoma,and explore their correlation with the occurrence,development,invasion,and metastasis of the tumor. Methods PV-6000 immunohistochemistry was used to measure the expressions of RECK and MMP-2 in 30 samples of osteosarcoma with complete clinicopathological data,and another 10 cases of osteochondroma served as a control. Results The expression rate of RECK in osteosarcoma was low（30.0%）,and that in osteochondroma was high（80.0%）,the difference between them being significant（χ2=5.76,P0.05）.The expression of MMP-2 in osteosarcoma was high（76.7%）,and that in osteochondroma was low（10.0%）,the difference between them was also significant（χ2=11.25,P0.05）.The expression of RECK was negatively correlated with that of MMP-2 in osteosarcoma（r=-0.84,P0.05）. Conclusion In osteosarcoma,the expression of RECK is low,and expression of MMP-2 is high.RECK and MMP-2 may be served as important markers to predict the infiltration and metastasis of this malignancy.
