壳寡糖对兔骨关节炎的治疗效果

目的 探讨关节腔内注射壳寡糖对兔骨关节炎的治疗效果及其作用机制.方法 新西兰兔12只,于右膝关节腔内各注射4 g/L的木瓜酶溶液0.3 mL,随机分为生理盐水对照组及20 g/L的壳寡糖实验组.自造模7 d后右膝关节开始给药,每周1次,共5周.于末次给药后7 d以空气栓塞法处死动物,关节软骨及滑膜组织以苏木精-伊红（HE）染色,光镜下观察其病理变化,免疫组织化学染色法检测膝关节软骨及滑膜组织肿瘤坏死因子α（TNF-α）的表达.结果 HE染色表明,实验组退变软骨及滑膜的病理变化轻于对照组,免疫组织化学染色法显示,实验组TNF-α的表达明显低于对照组.结论 壳寡糖能够明显降低骨关节炎滑膜、软骨组织中TNF-α的表达水平,延缓软骨的退变程度,可能成为一种新的骨关节炎治疗药物,值得进一步研究、开发和利用.

THE EFFECT OF CHITOSAN ON OSTEOARTHRITIS IN RABBITS

Objective To investigate the effect and mechanism of intraarticular injection of chitosan on osteoarthritis in rabbits. Methods A model of osteoarthritis was created in 12 New-Zealand rabbits by injecting 0.3mL of 4 g/L papain into ar- ticular cavity of the right knee. The rabbits were randomly divided into two group; chitosan experiment and normal-saline-control. Medication was started seven days after the creation of the models, once a week for five weeks. All rabbits were sacrificed by aeroembolism seven days after the last medication. The pathology of cartilage and synovium of the knee joint was examined by HE staining and expression of TNF-α detected by immunohistoehemistry. Results In comparison hetween the two groups, HE staining indicated that pathological changes of the cartilage and synovium were milder in the experiment group than the control; immunohis- tochemically, the expression levels of TNF-α were lower. Conclusion Chitosan oligosaceharide can obviously reduce the expressions of TNF-α in cartilage and synovium of osteoarthritis, delay the course of cataplasia of the cartilage, which can possibly become a new drug worth further research, development, and exploitation for the therapy of this condition.
