大鼠脑纹状体出血后神经功能损伤的研究

摘    要:目的:研究影响大鼠脑出血(ICH)后神经功能的因素。方法:将88只SD大鼠随机分为NCG、SCG、ICHG,各组根据术后时间又分为12h、1d、2d、3d、7d5个亚组;用胶原酶/肝素/生理盐水液注入大鼠脑纹状体制备ICH模型;分别在术前和术后各时间点对大鼠进行神经功能评分;干湿重法测脑水含量;TUNEL法观察凋亡细胞;电镜观察血肿周围细胞超微结构改变。结果:1ICH后大鼠出现不同程度神经功能损伤,在ICH3d神经功能损害最严重;2ICHG脑水含量明显高于SCG,以ICH3d脑水含量最高;3在ICH12h血肿周围出现TUNEL阳性细胞,3dTUNEL阳性细胞数最多,范围最广,以神经细胞为主;4电镜观察到ICHG血肿周围神经细胞出现凋亡细胞的形态改变。结论:ICH后血肿对周围神经组织的压迫、炎症反应、脑水肿以及细胞凋亡均为大鼠神经功能损伤的因素。

Study of neurological impairment after striatum hemorrhage in rats

Abstract:Objective: To explore the factors which affect the neurological impairment after intracerehral hemorrhage(ICH). Methods:88 healthy SD rats were randomly divided into three groups; NCG(n = 8), SCG(n= 40) and ICHG(n= 40). The SCG and ICHG were subdivided into 12h,1d, 2d, 3d, 7d five groups. Ⅵ Collagenase/heparin in saline solution or pure saline solution was injected into the corpus striatum. Neurological impairment was evaluated at 12h, 1d, 2d, 3d, 7d after the injection respectively. Brain water content was assayed by means of dry-wet weight method. TUNEL staining was employed for the frozen sections. The positive cells were observed by the microscope. Perspective electronic microscope was used to observe cell ultrastructure changes after ICH around hematoma. Results:Neurological impairment occurred in each rat after ICH, and the most severe neurological impairment was found at 3 d after injection. Brain' water content was the highest at 3 d after ICH. TUNEL positive cells were found at 12h, widespread at 3d after injection. The degree of the neurologic impairment was the same as that of the apoptosis. Conclusion: Apoptosis ,inflammatory and brain edema around hematoma may play important roles in neurological impairment after ICH.
