朝医麻黄定喘汤对哮喘模型小鼠肺组织中p38蛋白激酶表达的影响

目的]探讨朝医麻黄定喘汤对支气管哮喘模型小鼠肺组织中p38蛋白激酶(p38MAPK)表达的影响.方法]应用卵清蛋白腹腔注射致敏和反复超声雾化吸入方法刺激复制小鼠哮喘模型,随机分成正常对照组、哮喘模型组和麻黄定喘汤小、大剂量组.分别采用酶联免疫吸附法和蛋白质印迹法检测支气管肺泡灌洗液中的嗜酸性粒细胞总数(EOS)、白细胞介素5(IL-5)含量和肺组织磷酸化p38MAPK表达的变化.结果]朝医麻黄定喘汤小、大剂量组小鼠肺组织磷酸化p38MAPK表达水平、IL-5含量及EOS计数均较哮喘模型组显著降低(P<0.05),而且p38MAPK表达水平与IL-5含量和EOS计数呈正相关.结论]朝医麻黄定喘汤可能通过p38MAPK信号转导途径对哮喘起治疗作用.

Effects of the Mahuangdingchuan decoction of traditional Korean medicine on expression of p38 MAPK in asthma model of mice

OBJECTIVE To investigate the effects of Mahuangdingchuan decoction(MHDCD) from traditonal Korean medicine on expression of p38 mitogen-activated protein kinase(p38 MAPK) of lung tissue in bronchial asthma model of mice.METHODS The asthma models of mice were established by intraperitoneal injection with chicken ovalbumin and repeated ultrasonic atomizing inhalation,and the mice were randomly divided into 4 groups of control,asthma,small and large dose of the MHDCD.Total number of eosinophil(EOS),content of IL-5 in bronchoalveolar lavage fluid and expression of phosphorylated p38 MAPK of lung tissue were detected by using ELISA and Western blot.RESULTS The expression of phosphorylated p38 MAPK of lung tissue,total number of EOS and content of IL-5 were significantly lower in small and large dose groups of the MHDCD than in the asthama group(P<0.05),and the expression of phosphorylated p38 MAPK was positive correlation with total number of EOS and content of IL-5.CONCLUSION The MHDCD has a role of treating the asthma of mice via the activation of p38 MAPK signaling pathway.
