青蒿素联合琥珀酸亚铁对人肝癌细胞SMMC-7721选择性抑制作用的实验研究

摘    要:目的研究青蒿素联合琥珀酸亚铁对人肝癌细胞SMMC-7721的选择性抑制作用。方法将人肝癌细胞株SMMC-7721接种于小鼠腰部皮下,制成小鼠荷瘤动物模型,分别用青蒿素、琥珀酸亚铁、青蒿素+琥珀酸亚铁、生理盐水(对照组)对荷瘤小鼠进行干预,观察小鼠肿瘤体积的变化;TUNEL法检查肿瘤细胞的凋亡,计算抑瘤率和凋亡指数。结果青蒿素和青蒿素+琥珀酸亚铁与生理盐水相比,明显抑制荷瘤小鼠的肿瘤生长(P〈0.05),其抑瘤率分别为33.21%和55.04%,2组间存在明显差异(P〈0.05)。青蒿素联合琥珀酸亚铁表现出明显的促凋亡作用,凋亡指数为31.35%,与对照组相比差异有统计学意义(P〈0.05)。结论青蒿素和青蒿素联合琥珀酸亚铁对人肝癌细胞SMMC-7721均有肿瘤抑制作用和促进肿瘤细胞凋亡的作用,但是联合用药作用更强。

Experimental Study of the Selective Inhibition of Artemisinin Combined with Ferrous Saccinate toward Human Hepatoma SMMC-7721 Cells

Abstract:Objective To investigate the selective inhibition of artemisinin combined with ferrous succinate toward the human hepatoma SMMC-7721 cells. Methods The animal model of mouse-borne tumor was made by subcutaneous transplanting human hepatoma carcinoma SMMC- 7721 cells to the waist of mouse. The tumor bearing mice were intervened with artemisinin,ferrous suecinate, artemisinin plus ferrous succinate, physiological saline (control group), respectively,and gross tumor volume was observed. Apoptosis of tumor cell was detected by TUNEL,also tumor inhibition rate and apoptotic index were evaluated. Results Compared with physiological saline, artemisinin, artemisinin plus ferrous succinate obviously suppressed tumor growth of tumor-bearing mice(P〈0.05),the tumor inhibition rate of which was 33.21%, 55.04% respectively, which showed significant difference (P〈 0.05). Artemisinin combined with ferrous succinate greatly induced apoptosis of tumor cells,and apoptotic index of it was 31.35%,which statistical significfince compared with control group(P〈0.05). Conclusion Both artemisinin had and artemisinin combined with ferrous succinate had the effct on inhibition growth and promotion apoptosis of tumor cells,but the role of a stronger combination
