摘    要:目的研究青藤碱对食管癌EC109细胞增殖和凋亡的影响及其潜在机制。方法以不同浓度青藤碱分别处理体外培养的食管癌EC109细胞,72h后用CCK8法检测细胞增殖,用AnnexinV/PI双染与流式细胞术检测细胞凋亡,用Westernblot检测COX-2和Survivin蛋白的表达。结果采用0.2、0.4mmol/L青藤碱处理EC109细胞72h后,细胞增殖显著被抑制;细胞凋亡检测结果显示,与对照组相比,中、高剂量组细胞凋亡率显著升高;Westernblot检测结果显示COX-2和Survivin蛋白在中、高剂量组中表达水平显著下降。结论青藤碱在体外能抑制EC109细胞增殖并促进凋亡,其机制可能与抑制COX-2和Survivin蛋白表达相关。

Effect of Sinomenine on Proliferation and Apoptosis of EC109 Cell Through COX-2 Pathway

Abstract:Objective To observe the effect of sinomenine(SIN) on proliferation and apoptosis of EC109 cell and disclose the potential mechanism. Methods EC109 Cells were given different dose SIN. After incubated 72 h, cell proliferation and apoptosis were detected by CCK8 and Flow Cytometry, respectively. COX-2 and Survivin were investigated by Western blot. Results The proliferation of EC109 cell was inhibited after 72 h with 0. 2,0.4 mmol/L SIN. The apoptosis rate was increased in the experimental groups compared with that in the control group. The ex- pression of COX-2 and Survivin in the middle and high dose experimental groups were decreased compared with the control group. Conclusion SIN inhibits cell proliferation and enhances apoptosis in EC109 cells,in vitro, possibly by directly repressing COX-2 and Survivin expression.
