摘    要:以大肠杆菌HB101菌株为受体菌,研究了CaCl2浓度,转化时间及转化 温育时间对柯萨奇B组病毒3型cDNA重组质粒pGP51B转化作用的影响。实验结果表明,CaCl2浓度和转化时间对转化率的影响极为显著,而转化后的能时间对转化率的影响不明显。

Abstract:The effects of transformation conditions,which included CaCl2 concentration,transformation time and incubation time,on the transformation of recombinant plasmid pGP51B were investigated in this paper.pGP51B was a recombinant molecule of Coxsackie virus B3 cDNA fragment with the vector plasmid pGEM-1.The result showed that the effects of different CaCl2 concentration and transformation time on the transforn1ation frequency were significant differences,but the effect of incubation time after transformation was insignificant.The best condition for pGP51B transformation in this experiment was to prepare the competent host cells with 75mmol/L CaCl2 then transformating in ice bath for 30min following the direct selection on Amp+LB plate.
