慢性乙型肝炎肝郁脾虚证生物信息学差异研究

摘    要:目的:应用基因芯片技术研究慢性乙型肝炎肝郁脾虚证与它证的生物信息学差异。方法:应用Affymetrix Gene Chip人类全基因组芯片,分析了9例、3证型慢性乙型肝炎患者(肝肾阴虚型、湿热中阻型、肝郁脾虚型各3例)基因表达谱。通过应用Scanner 3000 7G 4C扫描仪对芯片进行扫描和信号值转换,获得各个中医证型之间的相对表达比值。结果:在检测的全部54676个基因中,中医证型基因表达存在着明显的差异,肝郁脾虚型与肝肾阴虚型、肝郁脾虚型与湿热中阻型比较,表达差异基因均占全基因的0.21%。结论:慢性乙型肝炎在肝郁脾虚证的基因层面上有着异常表达,这可能是临床出现同病异证的生物信息学基础,为其有效预防和治疗提供新的思路。

STUDY ON BIOINFORMATICS DIFFERENCES OF LIVER STAGNATION AND SPLEEN DEFICIENCY SYNDROME IN CHRONIC HEPATITIS B

Abstract:Objective:To study the bioinformatics differences between liver stagnation and spleen deficiency syndrome and the other syndromes in patients with chronic hepatitis B by gene chip technology.Methods:The gene expression profile of 9 cases with chronic hepatitis B(Yin deficiency of liver and kidney,damp-heat of liver and gallbladder,liver stagnation and spleen deficiency syndrome,3 cases respectively) were analyzed with affymetrix gene chip human genome chip.Results:There were obvious differences on gene expression in different syndromes which were differentiated based on TCM theory among 57676 kinds of genes that were obtained,to compare with Yin deficiency of liver and kidney syndrome and damp-heat of liver and gallbladderand,the different gene expression was 0.21% in total gene expression in liver stagnation and spleen deficiency syndrome.Conclusion:There were different gene expression in liver stagnation and spleen deficiency syndrome of chronic hepatitis B from the other syndromes,these provided new train of thoght for differentiation,prevention and treatment of liver stagnation and spleen deficiency syndrome of chronic hepatitis B,which were bioinformatics basis of different syndromes in the same disease,too.
