摘    要:目的:分析川芎嗪注射液直流电离子导入兔膝关节腔后,采用高效液相色谱法(HPLC)测定其含量以及有效成分的吸收、分布及代谢情况。方法:将川芎嗪注射液通过离子导入兔膝关节腔,选择5、15、30min不同的时间点抽取膝关节液,采用HPLC检测川芎嗪的含量,并计算其峰值与浓度的关系。结果:在1.20-35μg/mL范围内,膝关节液中川芎嗪平均回收率均在90%左右,日内精密度在5%左右,日间精密度在8%左右。结论:当川芎嗪浓度在一定范围内时,兔膝关节液中川芎嗪峰面积与浓度呈良好线性关系。

Abstract:Objective: To determine the content of ligustrazine in rabbit knee joint fluid with high performance liquid chromatography (HPLC) and to understand the absorption, distribution, metabolism and excretion of active component. Methods: Rabbit knee joint fluid was collected at different time (5,15 and 30 minutes) after ligustrazine injection direct current iontophoresis, and the content of ligustrazine was determined using HPLC, then relationship between the peak and concentration of ligustrazine was ana- lyzed. Results=In the range of 1. 20-35 μg/mL in rabbit knee joint fluid, the average recovery rate was 90%, RSD of intra-day was 5%, inter-day was 8%, respectively. Conclusions. The peak area has good lin- ear relationship with concentration of the ligustrazine in rabbit knee joint fluid in some invariably range.
