摘    要:目的建立一种简单快速的方法来测定岩黄连总生物碱含量,为岩黄连质量控制提供依据。方法采用氯仿超声提取岩黄连不同部位样品,依据酸性染料比色法的原理,以盐酸小檗碱为对照,溴甲酚绿为指示剂,在416 nm波长下比色测定样品中总生物碱含量。结果吸光度值与生物碱含量线性关系良好(r=0.999 1);回收率为101.91%,RSD为1.75%;测定结果在3 h内稳定,RSD为1.09%;岩黄连根部生物碱含量最高,达到48.77 mg/g,叶和夹果其次,花和茎中最低。结论该测定方法简便可靠,可用于岩黄连质量控制;部位不同总生物碱含量也不同。

Abstract:ObjectiveAn effective method was established in order to determine the total alkaloids from different parts of Corydalis saxicola Bunting.MethodsUsing ultrasonic cleaner,the total alkaloids were extracted from the samples and tested at 416 nm with the spectroscopy by the means of acid dye method.ResultsThe calibration curve of berberine were linear((r=0.999 1)),and the recovery rate was perfect(101.91%).The results of measurement were consistent in 3 hours.The content of alkaloids in roots was the highest,with 48.77 mg/g,followed by leaves/fruits and flowers/stems.ConclusionThis method is simple and effective,and the contents of total alkaloids were different among different parts of C.saxicola.
