基于UPLC特征图谱及两种成分含量测定的青翘质量评价*

摘    要:目的:建立中药青翘的超高效液相(UPLC)特征图谱并对2种活性成分进行含量测定,为青翘的质量评价提供依据。方法:利用UPLC法建立青翘特征图谱,通过相似度评价,结合聚类分析(CA)、主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)对12批不同产地和不同加工方式的青翘饮片进行质量评价。参照《中国药典》2020年版连翘含量测定项下测定连翘苷和连翘酯苷A的含量。结果:青翘UPLC特征图谱共确定了7个共有峰,其相似度均大于0.98;聚类分析和主成分分析将12批青翘样品划分为4类,在产地和加工方式上未呈现明显规律性;利用正交偏最小二乘法判别分析,确定了3个质量差异标志物,即峰5(连翘酯苷A)、峰2和峰4。含量测定结果显示,河南和山西产区的样品中连翘苷和连翘酯苷A的含量均符合《中国药典》的规定,河南西峡产区的样品中连翘苷和连翘酯苷A含量差异较大。结论:该方法可以用于评价不同产地青翘样品质量的差异,为青翘的综合利用提供了实验依据。

Abstract:Objective: To establish UPLC characteristic chromatograms and determine the contents of two active components of Fructus Forsythia for providing basis for the quality evaluation of Fructus Forsythia. Methods: UPLC method was used to establish the characteristic atlas of Fructus Forsythia, and the quality of 12 batches of Fructus Forsythia from different habitats and different processing methods was analyzed by similarity evaluation, combined with cluster analysis (CA), principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA). The contents of forsythin and forsythoside A were determined according to the determination of Fructus Forsythia content in the 2020 edition of Chinese Pharmacopoeia. Results: There were 7 common peaks in UPLC characteristic chromatogram of Fructus Forsythia, and the similarity was greater than 0.98; The 12 batches of Fructus Forsythia were basically classified into 4 groups by CA and PCA, and there was no obvious regularity in origin and processing method; Three quality difference markers were determined by OPLS-DA analysis, namely, chromatographic peak 5(forsytho‐side A), peak 2 and peak 4. The content determination results showed that the contents of forsythin and forsythoside A in the samples from Henan and Shanxi production areas were in line with the provisions of the Chinese Pharmacopoeia, and the contents of forsythin and forsythoside A in the samples from Xixia in Henan province were significantly different. Conclusion: This method can be used to evaluate the quality difference of the samples from different places of origin, and provide an experimental basis for the comprehensive utilization of Fructus Forsythia.
