Abstract:Objective The IDH1 R132H mutation and the expression of glioma cell line IDH1 in human brain glioma tissues of different levels were detected to investigate the effect of IDH1 R132H mutation on glioma of different levels.Methods IDH1 expression in glioma cell lines such as U87,U251,ln-229,HS683 and U118 was detected by q-PCR.Retrospective collection of 70 cases of glioma tissue specimens confirmed by surgical resection and pathology(experimental group)and 20 cases of brain tissue specimens obtained by intracranial decompression during the same period due to brain trauma(control group).IDH1 R132 H mutation was detected by immuno?histochemistry and western blot.The obtained data were pantwise compared by SNK method or K-W test,the categorical variables were tested byχ2 test,and the survival analysis compared the prognosis.Results IDH1 was relatively high in U87 and HS683,and relatively low in U251,U118 and ln-229.Seventy cases of the organization for human brain glioma specimens in 36 cases of mutations,mutation rate was 51.4%,the mutation occurs mainly inⅡ(51.7%),Ⅲ(79.2%)grade glioma and astrocyte tumors(80.0%),the difference is sta?tistically significant(P<0.05).The mutation of IDH1 R132H in glioma tissues of the experimental group was related to the age,epileptic seizure,pathological grade and Ki-67 expression of the patients(P<0.05).Ⅱ,Ⅲgliomas in IDH1 R132H prognosis is good(P<0.05),Ⅳlevel in patients with glioma IDH1 R132H no obvious correlation with prognosis(P=0.72).Conclusion IDH1 is relatively highly ex?pressed in U87 and HS683 cell lines,which provides a good cell model for the study of the pathogenesis of glioma.IDH1 R132H tend to occur inⅡ,Ⅲastrocyte tumors patients,and the mutation of IDH1 R132H was related to the patient’s age,epileptic seizure,pathological grade and Ki-67 expression.Mutant IDH1 R132HⅡ,Ⅲgrade gliomas patients have better prognosis than other patients who were not mutation.
