Abstract:Objective To establish an UPLC-MS/MS method for the determination of the concentrations of dezocine and ropivacaine in human plasma.Methods Venous blood samples were precipitated with acetonitrile for protein extraction,and processed with lidoca as an internal standard.The analytical column was packed with Agilent Zorbax Eclipse Plus C18.The mobile phase was 0.1%ammonium formate solution-methanol(75∶25,pH adjusted to 4.5 with formic acid)at a flow rate of 0.2 mL·min-1,the column temperature was 35℃and the injection volume was 2μL.The multi-reaction monitoring mode was used to quantitatively analyze with electrospray positive ionization.The blood concentration of dezocine and ropivacaine in the plasma of 30 hospitalized patients were determined by the newly established method.Results Excellent liner relationships of dezocine and ropivacaine were obtained in the ranges of 0.23-90.48 ng·mL-1 and 1.23-490.34 ng·mL-1,respectively,the limit determinations were 0.23 and 1.23 ng·mL-1.The average extraction recoveries were 90.11%-94.58%and 94.81%-101.33%.The inter-day and intra-day RSDs were less than 15%.Among 30 patients,the blood concentration ranges of dezocine and ropivacaine were 0.57-3.79 ng·mL-1 and 2.30-14.03 ng·mL-1,respectively.Conclusion The method is accurate,simple and rapid with high sensitivity,which can be used to analyze the clinical samples and determine the pharmacokinetics of dezocine and ropivacaine metabolites.
