紫外线对系统性红斑狼疮患者外周血T淋巴细胞Fas抗原表达的影响

摘    要:目的观察紫外线对系统性红斑狼疮(SLE)患者T淋巴细胞Fas抗原的影响。方法将30例SLE患者分为活动期组(A)和非活动期组(B),另设健康对照组(C)15人。采静脉血进行淋巴细胞分离及培养(其中一个培养板进行紫外线照射)后,用双标荧光法分析并获得CD+Fas+细胞百分数。结果在未经紫外线处理的A组Fas表达均显著高于B组(q=9.45,P<0.01)和C组(q=10.71,P<0.01)。B组及C组之间差异无统计学意义(q=1.10,P>0.05);而在经紫外线处理后的A组及B组Fas表达均明显上调(P<0.01)。同时,三组之间q检验显示q值分别为17.29,9.64和7.53,P值均小于0.01,说明均有统计学意义。结论SLE患者外周血T淋巴细胞在紫外线的刺激下更敏感地上调Fas抗原的表达。因此,很可能通过此途径进一步促进T淋巴细胞凋亡。

Abstract:Objective To investigated the effect of ultraviolet ray on Fas antigen and apoptosis of T lymphocytes in patients with systemic lupus erythematosus (SLE). Methods Thirty-six patients with SLE were divided into an active group and an inactive group according if they were in the active phase, and 15 normal subjects were recruited as the control group. Lymphocytes of every subject was isolated from their vein blood and cultured in two flasks. One of the flasks was irradiated by ultraviolet ray, while the other was not. Percentage of the CD_3+Fas+ cell was gained by double-fluorescence staining. Results In the flasks without ultraviolet ray irradiation, the level of Fas expression in active group was higher than that in inactive group (q=9.45, P<0.01) and that in control group (q=10.71, P<0.01), and no significant difference between inactive group and control group was revealed (q=~1.10 ,P>0.05). While in the flasks with ultraviolet ray irradiation, the level of Fas expression in both active and inactive groups elevated (P<0.01), which was significantly different from that of the control group. Conclusion Ultraviolet ray irradiation of the peripheral blood T lymphocytes of patients with SLE could up-regulate the Fas antigen sensitively. This could in turn induce the apoptosis of T lymphocytes.
