2-(2-苯并呋喃基)-2-咪唑啉对大鼠脑缺血后神经功能及血管新生的影响

摘    要:目的:观察2-(2-苯并呋喃基)-2-咪唑啉[2-(2-benzofu-ranyl)-2-imidazoline,2-BFI]对局灶性脑缺血大鼠神经神经功能恢复和缺血半暗带区新生血管形成的影响。方法:制备SD大鼠大脑中动脉阻塞(MCAO)模型,随机分为假手术组、2-BFI组和对照组,通过改良神经功能缺损评分(modified Neurological Severity Scores,mNSS)、TTC染色、免疫组化学检测脑内vWF的表达、异硫氰酸荧光素右旋糖酐(FITC-dextran)法行脑微血管灌注成像等研究方法研究2-BFI对神经功能、脑缺血梗死区的体积、内皮细胞及血管新生影响。结果:①2-BFI组的神经功能缺损评分(NSS)有明显改善(P<0.05);②2-BFI组脑梗死体积百分比小于对照组(P<0.05);③2-BFI组阳性细胞数明显高于对照组(P<0.05);④2-BFI组的脑内缺血半暗带区域毛细血管分布较为密集且充盈状态良好,微血管显影面积明显高于对照组(P<0.05)。结论:2-BFI能促进脑缺血大鼠神经功能恢复,促进脑内缺血半暗带区域血管新生,增加微血管血流灌注。

Abstract:ABSTRACT Objective: To study the effects of 2 - (2-benzofu ranyl)-2-imidazoline (2-BFI) on angiogenesis in ischemic boundary zones and neurologic function recovery in rats with focal cerebral infarction. Method:The rat model of middle cerebral artery occlusion (MCAO) was made. Neurobehavioral deficits, infarction volume, vWF expression, angiogenesis and cerebral microvessel perfusion were studied in the sham operation group, the 2-BFI group and the control group. Result:Compared with the control groups, the 2-BFI group had significantly improved the neurological score, reduced infarction, activated the expression of vWF and improved cerebral microvessel perfusion(P<0.05). Conclusion:2-BFI can stimulate angiogenesis, improve neurological function recovery and blood supply in brain ischemic boundary zones in SD rats.
