尿激酶型纤溶酶原激活剂单克隆抗体的制备及对癌症诊断的临床应用

目的：获得特异的鼠抗人尿激酶型纤溶酶原激活剂的单克隆抗体，以建立特异、灵敏、准确的癌症临床诊断方法。方法：以纯化的尿激酶型纤溶酶原激活剂（ｕＰＡ）为抗原免疫Ｂａｌｂ／ｃ小鼠，制备单克隆抗体。结果：利用小鼠杂交瘤技术，建立４ 株分泌ｕＰＡ 抗体的杂交瘤细胞系６Ｇ９、６Ｇ１０、５Ｅ１２、６Ｈ２，经鉴定其抗体类别均为ＩｇＧ，它们均能特异识别ｕＰＡ，与其它因子无交叉反应。结论：提示癌瘤组织中ｕＰＡ 含量对临床诊断及预测转移的可能性具有临床意义

Preparation of monoclonal antibodies against urokinase-type plasminogen activator (uPA) and clinical application in the diagnosis of cancer

Objective:To obtain specific McAb against human uPA and establish specific,sensitive,correct methods in the diagnosis of cancer.Methods:BALB/c mouse was immunized by purified uPA to prepare McAb.Results:Four positive hybridome cell lines which contained McAb against uPA were obtained.All McAb were IgG 1 according to the examination of immunoglobulin classification and they were named 6G 9,6G 10 5E 12 ,6H 12 .Western blot test showed that the four McAb reacted specifically with uPA without other crossing reactions.Conclusion:It is suggested that uPA contens in cancer tissues are clinically useful for diagnosing and predicting the metastatic of these cancers to the lymph nodes.
