PTEN和hMLH1蛋白的表达与胃癌发生及生物学行为的关系

目的探讨PTEN和hMLH1蛋白表达与胃癌发生及生物学行为的关系。方法免疫组化SP法检测80例胃腺癌细胞和68例非癌胃黏膜上皮细胞PTEN和hMLH1蛋白的表达情况,分析二者表达与胃癌生物学行为的关系。结果胃癌组PTEN蛋白的阳性表达率77.5%低于非癌胃黏膜组的91.2%(P<0.05),胃癌低分化组PTEN蛋白的阳性表达率61.1%低于高中分化组的88.6%(P<0.01);胃癌组hMLH1蛋白的阳性表达率67.5%高于非癌胃黏膜组的32.4%(P<0.01),低分化组hMLH1蛋白的阳性表达率50.0%低于高中分化组的81.8%(P<0.01),有淋巴结转移组hMLH1蛋白的阳性表达率85.7%高于无淋巴结转移组的57.7%(P<0.05)。胃癌细胞PTEN和hMLH1蛋白表达呈正相关(P<0.01)。结论PTEN蛋白表达下调可能与胃癌发生有关,而hMLH1在胃癌组的表达可能与代偿性修复有关。PTEN和hMLH1蛋白表达也会影响胃癌的部分生物学行为,其机制可能是hMLH1基因功能缺陷导致PTEN基因表达下调。

Expression of PTEN and hMLH1 in gastric carcinoma and their clinicopathological significance

Objective:To study the expression of PTEN, hMLH1 in gastric carcinoma and their correlations with occurrence, biological behavior. Methods :The expression of PTEN, hMLH1 was detected by immunohischemical method (s-p) in 80 specimens of gastric carcinoma cell and in 68 specimens of normal gastric mucosal epithelia cell .The expression of PTEN, hMLH1, and the relation with biological behavior was analyzed . Results: The positive expression of PTEN in two groups (gastric carcinoma, normal gastric mucosa) were 75%, 91.2% respectively, with the difference between two groups being statistically significant (P<0.05); Therates of hMLH1 were 67.5%,32.4% (P<0.01) in two groups, The rate of PTEN in the subgroup with low differentiation (61.1%) was higher than that in the group with high differentiation (88.6%) (P<0.01).The rate of hMLH1 were 50.0%,81.8% (P<0.01).The rate of hMLH1 in the subgroup with lymphatic metastasis (85.7%) was higher than that in the group without metastasis (57.7%) (P<0.05).The positive rate of PTEN expression was positive correlatedn with hMLH1 in gastric carcinoma cell (r=0.511,P<0.01) .Conclusion :The study indicated that down regulation of PTEN, hMLH1 correlated with the occurrence of gastric carcinoma, they possibly had affection on part of biological behavior. The mechanism is possibly that dysfunction of hMLH1 gene brig about the down regulation of PTEN gene expression.
