PDTC对重症急性胰腺炎E-selectin、氧化应激相关酶表达的影响

目的 探讨吡咯烷二硫代氨基甲酸脂(pyrrolidine dithiocarbamate,PDTC)对重症急性胰腺炎(severe acutepancreatitis,SAP)大鼠胰腺组织E.selectin和氧化应激相关酶的影响.方法 36只sD大鼠随机分成6组:正常组(n=6)、SAP组(n=6)、左旋精氨酸注射前1 h注射l mg/kg PDTC干预组(A组,n=6)、左旋精氨酸注射后3 h注射l ms/kgPDTC十预组(B组,n=6)、左旋精氨酸注射前1 h注射100 ms/kg PDTC干预组(C组,n=6)、左旋精氨酸注射后3 h注射100 mg/kg PDTC干预组(D组,n=6).采用腹腔内注射20%左旋精氨酸制作大鼠SAP模型.正常埘照组以生理盐水代替左旋精氨酸注射24 h后检测胰腺组织髓过氧化物酶(myeloperoxidase,MPO)、脂质过氧化物酶(1ipid peroxide,LPO)、一氧化氮合酶(nitric oxide synthase,NOS)的活性,免疫组化测定胰腺组织E-selectin,凝胶电泳迁移阻滞试验检测核转录因子NF-KB DNA结合活性.结果 在SAP组及SAP+PDTC组中,NF-KB、MPO、LPO、诱生型NOS(inducible NOS,iNOS)活性及E-selectin表达高于对照组(P<0.05),l mg/kg PDTC干预组(特别是模型诱导前1 h干预组)下调上述指标(P<0.05);结构型NOS(constitutive NOS,cNOS)活性低于对照组(P<0.05),而1 mg/kg PDTC干预组恢复cNOS活性(P<0.05).100 ms/kg PDTC对上述指标无影响.结论 1 mg/kg PDTC通过抑制NF-KB减轻SAP炎症程度,而100 ms/kgPDTC则无作用.

Effect of pyrrolidine dithiocarbamate on expression of E-selectin and oxidative stress related enzymes in rats with severe acute pancreatitis induced by L-arginine
