摘    要:目的:建立HPLC法测定不同来源补骨脂中补骨脂素和异补骨脂素含量的方法。方法:选用Hypersil C18柱(250mm×4.6mm,5μm),以甲醇:0.015%磷酸溶液(40:60)为流动相,流速1.0ml·min^-1,检测波长246/1/11,柱温25℃。结果:补骨脂素在4.16~14.56μg·ml^-1间呈良好的线性关系(r=0.9997),平均加样回收率为99.9%(RSD=0.90%,n=9);异补骨脂素在4.16~14.56μg·ml^-1间呈良好的线性关系(r=0.9996),平均加样回收率为98.0%(RSD=0.78%,n=9)。结论:该方法快速、准确,可用于补骨脂的质量控制。

Abstract:Objective: To establish an HPLC method for the simultaneous determination of psoralen and isopsoralen in psoralea. Method: The analysis was performed on a column of Hypersil C18(250 mm ×4. 6 mm ,5 μm). The mobile phase consisted of methanol and 0. 015% H3PO4 (40: 60) with the flow rate of 1.0 ml·min^-1. The detection wavelength was 246 nm and the column temperature was 25℃. Result: Promising linearity was obtained within the range of 4. 16-14. 56 μg·ml^-1 for psoralen and isopsoralen. The average recovery of psoralen and isopsoralen was 99. 9% (RSD = 0. 90%, n = 9) and 98.0% ( RSD = 0. 78% , n = 9 ), respectively. Conclusion: This method is rapid and accurate and can be used to control the quality of psoralea.
