雄黄对人结肠癌LoVo细胞生长及凋亡的作用

 目的 研究雄黄对结肠癌LoVo细胞生长及凋亡的调控作用及其机制。方法 用雄黄作用于体外培养的人结肠癌LoVo细胞，分别用细胞计数和MTT（四甲基偶氮唑蓝）比色法分析其对LoVo细胞生长的抑制作用，采用流式细胞仪测定细胞周期分布和凋亡率，用光镜观察细胞形态。结果 雄黄在（2.5～160.0）μg／ml质量浓度范围内呈剂量依赖方式抑制结肠癌LoVo细胞生长（P＜0.05），雄黄（12.5～50.0）μg／ml作用24 h后，光镜可见LoVo细胞呈典型的凋亡形态学改变；流式细胞仪检测出现凋亡峰，其测得的细胞凋亡率与对照组相比差异有统计学意义（P＜0.05）。结论 雄黄对人结肠癌LoVo细胞生长具有抑制作用，其机制与诱导肿瘤细胞凋亡有关，对细胞周期的影响表现为G2／M阻滞。

The effect of realgar on growth and apoptosis in human colon carcinoma LoVo cells

Objective To investigate the effect of realgar on growth and apoptosis in human colon carcinoma cell line LoVo. Method In vitro cultured human colon carcinoma cell line LoVo was treated with realgar, Cell proliferation was evaluated by MTT and cell acount assay. Distribution of cell cycle and apoptosis rate were determined by flow cytometry,and apoptosis was confirmed by morphology. Results The prolifera－tion of LoVo was inhibited significantly by realgar in a dosage dependant manner(range from 2.5 to 160.0 μg/ml, P ＜0.05). Observing under microscope, some LoVo cells underwent a typical apoptosis after incubation with re－algar for 24 h. Cells apoptosis rate was significantly increased in realgar group compared with that in the con－trols (P ＜0.05). Conclusions The results indicated that realgar inhibits the proliferation of colon carcinoma LoVo cells. The anticancer mechanism is related to induce cells apoptosis and arrest cell cycle at G2/M phase.
