超深低温处理的异体肌腱移植

摘    要:目的建立一种新的保留人体异体肌腱活性的超深低温处理技术,临床应用后长期随访其疗效。方法将新鲜人体肌腱置入由10%二甲基亚砜及纯小牛血清组成的冷冻保护液中,在室温下平衡后置入液氮罐气液面6h,再浸入液氮中贮存不少于3个月。临床应用前,用40℃水浴快速复苏及洗涤去除冷冻保护剂,复苏肌腱作腱细胞活力测定。临床应用11例共修复指(趾)伸、屈肌腱26根。结果超深低温处理的腱细胞活力为(78.7±6.3)%,(±s,下同)。临床应用后异体肌腱无排斥反应,术后3根肌腱粘连明显。结论超深低温处理的肌腱是活体肌腱。临床应用效果满意,可推广使用。

Abstract:Objective To establish a new ultra - deep - low - temperaturetechnique that can maintain the activity of human allografting tendon and provide the tendons for clinical application. Methods The fresh donor tendon was preserved in the cryopreserved solution which included 10% dimethyl sulfoxide and fetal calf serum. Balanced at room temperature, the tendon was dropped into liquid nitrogen freezer for 6 hours and immersed no less than 3 months. It was quickly thawed and washed at 40℃temperature water and detached cryopreservation agent before use. Then the resuscitated tenocyte activity was examined .26 finger flexion and extension tendons in 11 cases were repaired. Results The tenocyte cell activity handled by ultra - deep - low – temperature was (78.7 ± 6.3)%.No rejection occurred after tendon allografting. Obvious adhesion was found in 3 cases after operation. Conclusions Tendon handled by ultra - deep - low - temperature was active. It can be widely propagated due to its satisfactory treatment outcome.
