摘    要:目的探讨14-3-3蛋白过表达对神经母细胞瘤细胞株(SH-SY5Y)的保护作用和可能机制。方法利用脂质体转染的方法在SH-SY5Y细胞株中瞬时表达14-3-3蛋白。通过四甲基偶氮唑盐(MTT)法、Ho-echst 33342染色法、流式细胞仪和荧光显微镜分别检测14-3-3蛋白过表达对神经母细胞瘤细胞株SH-SY5Y的细胞活性、细胞凋亡、细胞内活性氧类物质(ROS)水平的影响。结果 14-3-3蛋白过表达可促进SH-SY5Y细胞的增殖;抑制SH-SY5Y细胞内ROS的产生。结论 14-3-3过表达对SH-SY5Y起神经保护作用,其机制可能是通过抑制细胞内ROS的产生而实现的。

The protective effects of 14-3-3 overexpression on SH-SY5Y cells

Abstract:Objective To investigate the protective effects of the 14-3-3 protein overexpression in neuroblastoma cell line SH-SY5Y and its mechanisms.Methods 14-3-3 protein was transiently expressed in SH-SY5Y cells with Lipotap.The effects of 14-3-3 expression on cell viability,apoptosis and intracellular ROS level were measured by MTT assay,Hoechst 33342 staining and flow cytometry analysis,respectively.Results MTT assay and flow cytometry analysis shows that the overexpression of 14-3-3 protein increases the cell viability,and inhibits the production of ROS.Conclusion The overexpression of 14-3-3 protein in SH-SY5Y cells plays a neuroprotective role,which is achieved by promoting the proliferation of SH-SY5Y cells and inhibiting the production of intracellular ROS.
