黄芪皂苷在大鼠血管钙化中作用及可能机制

摘    要:摘要 目的:在大鼠血管钙化模型上用黄芪皂苷注射液腹腔注射观察对血管钙化的影响,并探讨黄芪皂苷在血管钙化中作用及可能机制。方法:采用维生素D3和尼古丁制备大鼠血管钙化模型;运用von Kossa染色、原子吸收测定钙含量及碱性磷酸酶(ALP)活性测定判断血管钙化程度;用生物化学方法测定血管一氧化氮、超氧化物歧化酶和丙二醛含量;原位缺口末端标记法检测血管平滑肌细胞凋亡。结果:VDN处理组大鼠血管von Kossa染色见平滑肌细胞内和细胞间基质有大量棕黑色颗粒聚集;主动脉钙含量和ALP活性分别较对照高3.6和1.7倍(P<0.01),血管钙化后可加重血管组织氧化性损伤,增加血管平滑肌细胞凋亡;而诱导钙化后注射黄芪皂苷明显改善上述指标,减轻血管钙化程度和组织氧化损伤,平滑肌细胞凋亡细胞数量明显减少。结论:黄芪皂苷主要通过抑制血管平滑肌细胞氧化应激和细胞凋亡而减轻血管钙化。

Abstract:ABSTRACT objective To observe the effect of astragalus saponins on vascular calcification in rat, to explore the role and the possible mechanism of astragalus saponins in vascular calcification. Methods Vascular calcification model in rats was induced by administration of vitamin D3 plus nicotine. To estimate the extent of calcification by Von Kossa staining,calcium content and alkaline phosphatase activity. The nitric oxide(NO) content and activities of superoxide dismutase(SOD) and malondialdehyde(MDA)were measured by biochemistry. Vascular apoptosis was detected with in TUNEL staining.Result A strong positive staining of black/brown areas among the elastic fibers of the medial layer in calcified aorta by Von Kossa staining,calcium content and ALP activity in calcified arteries increased by 3.6-and 1.7-fold as compared with the controls;The NO content and SOD activity was greatly decreased 69% and 73%(P<0101);The lipid peroxidation products MDA in vascular were increased 85% (P<0.01); The apoptosis rate of vascular was significantly increase,compared with controls.However,calcium content and ALP activity in VDN plus astragalus saponins were lower than those in VDN group. Astragalus saponins alleviate vascular oxidative stress and relieve cell apoptosis.Condusion Astragalus saponins inhibit vascular calcification by decreasing vascular injury and reducing cell apoptosis
