葛花提取物对急性心肌梗死小鼠心肌的保护作用

摘    要:目的:阐明葛花对急性心肌梗死小鼠心肌的保护作用及作用机制。方法:结扎小鼠冠状动脉左前降支建立急性心肌梗死模型。实验分为生理盐水组,复方丹参组,葛花提取物低、中、高剂量组共5组。通过心肌梗死面积和心肌酶的变化来评价保护效果,通过检测血清中SOD、MDA的含量和心肌中β-ARK1、eNOS基因的表达情况来阐明其作用机制。结果:与生理盐水组比较,葛花提取物能够减少心肌梗死面积(P0.05),降低血清中心肌酶的含量(P0.05),同时能够升高血清中SOD的含量(P0.05),并能降低血清中MDA的含量(P0.05)和心肌中β-ARK1的表达(P0.05)。结论:葛花提取物通过升高SOD的含量和降低心肌中βARK1的表达来保护急性心肌梗死心肌。

Abstract:Objective To Elucidate the protective effect and active mechanism of Flos pueraria on myocardium of mouse with acute myocardial infarction. Methods Acute myocardial infarction mouse model was established by ligating the left anterior descending coronary artery. The experiment was designed into five groups include normal saline group, compound salvia miltiorrhiza group, Flos pueraria low-dose group, Flos pueraria mid-dose group and Flos pueraria high-dose group. The protective effect was evaluated by detecting myocardial infarct size and changes of myocardial enzymes. The active mechanism was elucidated by detecting the contents of serum SOD and MDA and the expressions of β-ARK1, eNOS in myocardium. Results Compared with the normal saline group, Flos pueraria could reduce the myocardial infarct size, decrease the content of serum myocardial enzymes, and increase the content of serum SOD, reduce the content of serum MDA and the β-ARK1 expression in myocardium. Conclusion Flos pueraria protects the myocardium of acute myocardial infarction mouse by increase the serum SOD content and decrease the β-ARK1 expression in myocardium.
