血吸虫病肝纤维化中碱性成纤维细胞生长因子的免疫组化研究

目的　研究血吸虫病肝纤维化时碱性成纤维细胞生长因子 (bFGF)在肝脏的表达情况及初步探讨其与纤维化的关系。方法　采用经皮肤感染日本血吸虫尾蚴法建立小鼠血吸虫病肝纤维化模型。利用免疫组化技术研究bFGF在肝纤维化过程中的表达情况 ,并用VG染色显示胶原纤维的沉积情况。结果　小鼠感染后 6周时 ,出现急性血吸虫卵肉芽肿 ,肝内bFGF阳性细胞的分布与对照组无明显差异 ,VG染色显示肉芽肿内无胶原沉积 ;8周时 ,许多bFGF阳性细胞聚集围绕在虫卵肉芽肿周围 ,且细胞形状由原来的多角形变为长梭形。同时 ,VG染色显示肉芽肿周围出现一些胶原纤维 :1 0周时 ,bFGF阳性细胞和胶原纤维均增多。结论　bFGF与血吸虫病肝纤维化有密切关系 ,可能在该病的发生和发展过程中起重要作用

Immunohistochemical study on basic fibroblast growth factor in schistosomal hepatic fibrosis

Objective To investigate the expression of basic fibroblast growth factor(bFGF)in schistosomiasis with liver fibrosis and the relation between bFGF and liver fibrosis.Methods Murine schistosomal hepatic fibrosis model was builded by making the mice infected with S.japonicum cercariae.Immunohistochemical method was used to observe the expression of bFGF in the course of liver fibrosis,and Van Giesons stainning was applied to show collagen fibrosis.Results At the 6th week after infection,acute egg induced granuloma formed.There was no obvious difference between the distribution of bFGF postive cells in the livers of model group and that of control group.Moreover,Van Giesons stainning showed that no collagen deposited in the liver granulomas.At the 8th week,many bFGF positive cells gathered and embraced the granulomas,and their shapes changed from polygon to elongation.At the same time,Van Giesons stainning showed that some collagen fibrosis apeared around the granulomas.At the 10th week,both the number of bFGF positive cells and the amount of collagen fibrosis were increased.Conclusion bFGF correlates with schistosomal hepatic fibrosis closely and maybe play an important role in the pathogenesis and development of the disease.
