褪黑素的抗自由基作用

摘    要:目的 考察褪黑素(MT)的抗自由基作用。方法 采用体内和体外两种实验方法,以过氧化脂质-丙二醛(MDA)含量为 指标,研究了MT的抗自由基作用。结果 在体外,MT(1.0,10 mmol/L)均能极显著降低过氧化氢和羟自由基所致大鼠脑匀 浆MDA的升高。体内实验,MT(1.0,10 mg/kg,ip,12 d)均可显著抑制四氧嘧啶所致小鼠脑组织MDA含量的升高(P<0. 05);而MT(1.0,10 mg/kg,sc,2 w)均能极显著的降低12月龄小鼠肝、脑组织MDA含量。结论 褪黑素可通过抗自由基作 用保护机体免受过氧化损伤,延缓衰老。这种作用可能是MT抗衰老的机制之一。

Abstract:Objective To study the anti-radical effect of Melatonin( MT) . Method The level of MDA was studied in vitro and in vivo in order to investigated the anti-radical action of MT. Result In vitro, MT(1.0,10 mmol/L) significantly relieved the content of MDA of rats brain homogentes which were induced by hydrogen peroxide or hydroxyl radical. In vivo, MT(1.0,10 mg/kg,ip,12d) could inhibit the increasing content of MDA in mice brain induced by alloxan; MT( 1.0,10 mg/kg,sc,2w)also could relieve the level of MDA in 12-month old mice brain and liver. Conclusion It was suggested that MT could inhibit the oxidative damage of radical by anti- radical effect. It is considered that this effect of MT is likely to be one of its anti-aging mechanisms.
