摘    要:目的 阐述As2 O3 (三氧化二砷 )治疗人红白血病机制。方法 采用人红白血病多药耐药株K562 / ADM作为实验模型 ,研究As2 O3 对该细胞MDR逆转的可能性。结果 ①K562 / ADM 细胞对ADM具有明显的抗性 ,与K562 细胞相比 ,抗性倍数约为 4 0倍 ,而两者对As2 O3 的IC50 接近 ,无显著差异 ;②低毒剂量的As2 O3 (1 0 μmol/L)与ADM(4 0 μg/ml)联合运用 ,可升高细胞内ADM的浓度 ,降低细胞对ADM的IC50 ,使该细胞对ADM的抗性有数倍逆转。结论 As2 O3 是一种有效的MDR逆转剂。

Abstract:Objective To illustrate mechanisms of arsenic trioxide (As 2O 3) in the treatment of K 562 / ADM cells. Methods We study the reversal effect of As 2O 3 on K 562 / ADM MDR cells in experiment model of rat. Results ① The resistance of K 562 / ADM cell line to ADM was 40 times than that of K 562 cell line, but the IC 50 value of both cell lines to As 2O 3 was similar (P>0.05); ② low cytotoxic dose (2.0 μmol/L) of As 2O 3 in combination with ADM (4.0 μg/ml) increased the intracellular ADM concentration (P<0.05) and declined IC 50 value of K 562 / ADM cell line to ADM by 2.5 times than the control. Conclusion As 2O 3 might be an effective anti tumor drug as well as an effective drug for reversal of MDR.
