2型糖尿病大鼠血清及脑组织β淀粉样蛋白水平研究

摘    要:目的:探讨2型糖尿病(2型DM)大鼠血清及脑组织β淀粉样蛋白(Aβ)水平的变化与发生阿尔茨海默病(Alzheimer’s Disease,AD)的相互关系。方法:高脂、高热卡饲料喂养+用小剂量链脲佐菌素(STZ)腹腔注射的方法复制2型DM大鼠模型,并与对照组同步实验。造模成功后2周分离大鼠血清及脑组织,分别测定Aβ,空腹血糖(FBG),空腹胰岛素(FINS)水平。结果:糖尿病组血清Aβ、FBG、FINS水平均显著高于对照组,脑组织Aβ水平显著高于对照组。结果:糖尿病组血清Aβ水平显著高于对照组,脑组织Aβ也明显升高。结论:患2型DM时,血清和脑组织Aβ明显增高,表明2型DM大鼠血清和脑组织Aβ升高可能与其并发AD有一定的关系。

The Study of Expression of β-amyloid of Serum and Brain Tissue in Type-2 Diabetic Rat

Abstract:Objective To investigate the inherent relation betweenβ -amyloid protein(Aβ )and Alzheimer disease(AD) in rats with type-2 diabetic mellitus (DM).Methods Type-2 diabetic rats were induced by high-fat and high-caloric diet and a very small dose of streptozotocin(STZ) through intravenous injection,and matched controls were experienced in the same time. Serum and brain tissue of rats were collected after the model of DM had been completed for two weeks. Aβ, FBG,FINS were determined.ReSultS The serum level of Aβ,FBG,FINS in DM group was significantly higher than that of the control group. The level of Aβ in the brain tissue was also evidently increased.Conclusion The serum level of Aβ was significantly increased and the level of Aβ in the brain tissue was increased in rats with DM,which may be related to the development of AD.
