人工合成HDV抗原27肽的应用—血清抗HDV的检测

采用自行设计、选择含天然ＨＤＡｇ部分片段的人工合成２７肽建立ＥＩＡ检测抗ＨＤＶ方法，在国内８家医院检测各类人员中血清抗ＨＤＶ状况。ＨＤＶ抗原合成肽由中国科学院上海生物化学研究所合成。结果：献血员（３１５例），甲型肝炎（１１３例）和非乙型肝炎（１５４例）中２例阳性；在ＨＢＶ感染者中共检出抗ＨＤｖ（＋）为２ｌ５／１４８２（１４．５１％）。其中ＡｓＣ为１４／４１４（３．３８％）、ＡＨ为１１／７６（１４．４７％），ＣＰＨ为５／２８（１７．８６％）、ＣＡＨ为３２／１８１（１７．６８％）ＳＨ为１７／８６（１９．７７％），ＬＣ为５２／１４２（３６．６２％），ＰＨＣ为３０／１３０（２３．０８％），另５４／４２５（１２．７１％）未分型。同一单位抗ＨＤＶ的检出与已往用市售试剂结果基本一致，与Ａｂｂｏｔｔ试剂符合率为９４．９％（７４／７８）。提示本ＨＤＶ抗原２７肽具有ＨＤＶ抗原活性，能特异地为天然抗ＨＤＶ识别，可作为检测抗ＨＤＶ诊断试剂。

A SYNTHETIC 27 OLIGOPETIDE OF HDV ANTIGEN AND ITS USE--DETERMI- NATION OF SERUM ANTI HDV IN DIFERENT POPULATION

A synthetic HDAg 27 oligopeptide , which was selected and designed by the authors andsynthesized by Shanghai Institute of Biochemistry , Chinese Academy of Science was used with EIAmethod to detect serum anti HD in different population at eight hospitals in China. Anti HD was positivein two of 315( 0.65%)blood donors and was negative in all of 113 cases of hepatitis A and 154 cases ofhepatitis non-B.Anti HD was positive in 215 out of 1482 cases of HBVM positive(14. 51%), among themanti HD was positive in 3.38%(14/414) of ASC, 14. 47%(11/76 ) of AH, 17. 86%(5/28)of CPH,17. 68%(32/181) ofCAH, 19. 77%(17/86) of SH, 36. 62%(52/142)of LC, 23.08%(30/130) of PHC,and 12. 71%(54/425) of non- typed group, respectively. These results were in coincidence with the previ-ous repotrs from the same unit. The detection rates of anti HD were different in different areas. The coin-cidence rate was 94.9%(74/78 ) when compared with Abbott EIA kit. The results suggested that theHDAg 27 peptide possessed the activity of the natural HDAg,and it could be recognized by natural ant- HD. Therefore, it may be used as a diagnostic agent for detecting anti HD.
