摘    要:目的了解转录因子E2F1基因敲除对造血干、祖细胞的影响。方法用不同抗体标记E2F1基因敲除及同窝野生对照小鼠的外周血、脾脏、骨髓等细胞,应用流式细胞仪进行分析,比较两组之间的差异。结果对检测结果进行统计分析发现,与野生型小鼠相比,E2F1基因敲除小鼠的外周血、骨髓细胞均有改变,脾脏细胞未见明显变化,骨髓前体B细胞、髓系祖细胞、造血干细胞等均有显著变化。骨髓造血干、祖细胞减少,G1期造血干细胞减少。结论 E2F1基因敲除可使小鼠造血干、祖细胞减少。

Abstract:Objective To study the influence of E2F1 gene knockout on the Hematopoietic stem cells and progenitor cells. Methods Cells from the peripheral blood, spleen and bone marrow of E2F1 knockout and wildtype mice were stained with kinds of antibodies, and analyzed by flow cytometry. Results Compared with wildtype mice, significant differences were found in the peripheral blood and bone marrow, pre B cell, myeloid progenitor cell and Hematopoietic stem cell in the E2F1 nockout mice. No. of Hematopoietic stem cell and progenitor cell decreased, and percent of G1 stage Hematopoietic stem cell decreased. Conclusion Hematopoietic stem cell and progenitor cell decreased in E2F1 gene knockout mice.
