糖尿病大鼠肾小球晚期糖基化终末产物对肾脏改变的影响

摘    要:目的:探讨晚期糖基化终末产物(AGE)在糖尿病肾病发生与发展中的作用。方法:采用ELISA方法检测STZ-糖尿病大鼠肾小球组织晚期糖基化终末产物(GTE-AGE)含量,同时检测尿蛋白/肌酐(Pr/Cr)比值及肾小球基底膜厚度(GBMT)。结果:糖尿病大鼠在病程12周时GTE-AGE已明显增加(P<0.01),20周时尿Pr/Cr比值开始升高(P<0.05),28周时GBMT明显增加(P<0.01);胰岛素治疗组,GTE-AGE含量、尿Pr/Cr比值和GBMT也明显增加(P<0.05),但增加速度减慢;胰岛素与所基胍治疗组到28周时仍与正常对照无显著差异。结论:AGE在肾小球内的大量形成和堆积与肾小球结核和功能的异常改变直接有关,是高血糖引起肾脏病变的一个重要环节。

Abstract:Objective To study the role of advanced glycation end products (AGE) in development and progression of diabetic nephropathy.Methods Content of GTE AGE in streptozotocin diabetic rats was measured with competitive enzyme linked immunosorbent assay (ELISA).Meanwhile,urine Pr/Cr ratio and glomerular basement membrance thickness (GBMT) were also detected.Results In group D,GTE AGE content was significantly increased at 12 week,and urine Pr/Cr ratio at 20 week.GBMT had been significantly increased at 28 week.In group DI,GTE AGE content,urine Pr/Cr ratio and GBMT were also significantly increased,but more slowly than in group D.There was not significant difference between group DIA and group C at 28 week yet.Conclusion The formation and accumulation of AGE in glomeruli is directly related to abnormal alterations in structure and function of glomeruli and is an important link in development of nephropathy by hyperglycemia.
