二苯乙烯类化合物E1对2型糖尿病大鼠血清游离脂肪酸组成的影响

摘    要:目的 以血清游离脂肪酸组成的变化反映糖尿病病情 ,探讨二苯乙烯类化合物E1对 2型糖尿病的影响。方法 iv小剂量链佐星 (2 5mg·kg- 1)损伤大鼠胰岛细胞造成其糖耐量异常 ,继而喂以高糖 高脂饲料诱发动物肥胖形成 2型糖尿病大鼠模型 ;采用气质联用色谱法 ,对 2型糖尿病大鼠血清游离脂肪酸谱进行测定。结果  2型糖尿病模型组大鼠血清游离脂肪酸谱组成中花生四烯酸含量明显低于正常对照组 ,给予二苯乙烯类化合物E1(5 0 0mg·kg- 1·d- 1,ig ,14d)治疗后的大鼠血清游离脂肪酸谱中花生四烯酸含量明显增加。二苯乙烯类化合物E1对豆蔻酸甲酯 ,棕榈酸甲酯 ,棕榈油酸甲酯 ,硬脂酸甲酯 ,油酸甲酯 ,亚油酸甲酯含量无明显影响。结论二苯乙烯类化合物E1对 2型糖尿病大鼠的治疗作用可能是通过增加游离脂肪酸谱中花生四烯酸含量来实现的

Abstract:AIM To investigate the effect of stilbenoid E1 on the development of diabetes mellitus reflected by the changes in serum free fatty acids composition. METHODS ]The experiment was performed on a rat model of type- 2 diabetes which was established by tail iv of a low dose of freshly prepared streptozocin(STZ, 25 mg·kg-1) which can injure the islet cells and cause impairment of glucose tolerance. Then, the rats were fed with high glucose-high fat diets in order to make them obese. At the same time, the effect of stilbenoid E1 on serum free fatty acid composition in type-2 diabetic rats was determined by gas-chromatography/mass-spectrometry. RESULTS The serum level of arachidonic acids among free fatty acids was significantly lower in type-2 diabetic rats than that in the controls, however, it was markedly increased in type-2 diabetic rats administered with stilbenoid E1(500 mg·kg-1·d-1, ig, 14 d). Stilbenoid E1 had no effect on contents of methyl myristate, methyl palmitate, methyl palmitolate, methyl stearate, methyl oleate and methyl linoleate. CONCLUSION 〗Stilbenoid E1 has a therapeutic effect on rats of type- 2 diabetes, possibly by means of increasing the level of arachidonic acids in serum.
