高侵袭性胰腺癌Panc-1细胞系中干细胞筛选结果分析

摘    要:目的探讨高侵袭性胰腺癌细胞中是否有比例较高的胰腺癌干细胞。方法采用Transwell小室肿瘤侵袭模型筛选、分离出胰腺癌细胞系Pane-1中的高侵袭性癌细胞(Pane-1-H)和低侵袭性癌细胞(Pane-1-L),FCM检测上述两种细胞的胰腺癌干细胞表面标记物CD34、CD44、ESA。结果Pane-1-H、Pane-1-L中胰腺癌干细胞所占比例相近,随着侵袭时间延长,胰腺癌干细胞比例无改变,P均〉0.05。结论高侵袭性胰腺癌细胞中不含有较高比例的胰腺癌干细胞。

Abstract:Objective To study whether there is more pancreatic cancer stem cells in high-invasive pancreatic cancer cells. Methods The transwell cave tumor invasion model was applied to select high-invasive cancer cells and lower-invasive cancer cells in the Panc-1 pancreatic cancer cell lines. FCM was used to detect the expression of pancreatic cancer stem cell surface markers CD24, CD44, ESA in high-invasive pancreatic cancer cells and lower-invasive pancreatic cancer cells. Result The proportion of pancreatic cancer stem cells showed no differences in high-invasive pancreatic cancer cells and lower-invasive pancreatic cancer cells and did not change with the prolongation of invasion time, P 〉 0.05. Conclusion there is no more pancreatic cancer stem cells in high-invasive pancreatic cancer cells.
