诱导性人工骨膜的研制及实验研究

摘    要:目的 本研究研制了一种诱导性人工骨膜。方法 用猪真皮提取纯化胶原,盐析法制备胶原膜,并用此吸附BMP,制备成胶原-BMP人工骨膜。用大鼠桡骨3mm长骨缺损作为骨折模型。设空白、单纯胶原膜、胶原-BMP膜包绕骨折段以及新鲜同种异体植骨共4组。术后0.5、1、2和3个月分批处死各组大鼠数只,处死前2天每只大鼠腹腔内注射钙黄绿素80mg/kg。处死后分别行X线和软X线摄片,血清BGP测定以及荧光显微镜观察。结果 胶原-BMP人工骨膜组的上述多项检测指标均表明其成骨效能明显优于其他各组。结论 作者认为该组成骨效果优良的原因是由于胶原膜吸附了BMP,同时具有骨诱导和骨引导的两种成骨作用,而仅胶原膜包绕骨折段的单纯骨引导效果并不理想。

Abstract:Objective in this study, a kind of inductive artificial periosteum was prepared. MethodsThe collagen of porcine dermis was separated and purified, and the collagen membrane was made with themethod of the salt precipitation, and mixed with bone morphogenetic protein (BMP) to prepare artificial pe-riosteum containing collagen and BMP. A defect of 3 mm in length was produced at the radius of rats as fracturemodel, the rats were divided into four groups including control, collagen membrane, collagen-BMP membraneand fresh allograft These two kinds of membrane were respectively used to enclose the fractures. The rats wereseparately euthanized at 0. 5, l, 2 and 3 months postoperatively and the serum were collected- Two days beforesacrifice, each rat was given intraperitoneal injection of calcein at the dose of 80 mg/kg. X-ray and soft X-rayradiography of right forearm were taken and the level of BGP in serum was measured. The histologic changes ofthe samples were observed under fluorescent microscope. Results The results showed that osteogenic abilityin collagen-BMP membrane group was superior to that of other groups. Conclusion The collagen-BMPmembrane had the role of osteoinduction as well as guided bone regeneration.
