低氧对鼠雪旺氏细胞增殖的影响

目的观察低氧对鼠雪旺氏细胞体外增殖的影响,为雪旺氏细胞扩增培养探索新方法。方法原代分离培养雪旺氏细胞并鉴定,将培养至第三代的细胞分为3组,分别为对照组,10%02组和3%O2组。对各组细胞记数;测定培养液中氧含量和乳酸浓度及各组细胞的增殖指数。结果随着环境氧浓度的下降,细胞培养液中的氧含量也逐渐下降(P<0.05),而乳酸浓度逐渐升高(P<0.05)。低氧组雪旺氏细胞数目比常氧组明显增加,尤以10%O2组更为明显(P<0.05),3%O2组在低氧1d和2d时增殖指数低于常氧组,低氧4时,增殖指数增加;而10%O2组增殖指数一直高于常氧组(P<0.05)。结论轻中度低氧有利于鼠雪旺氏细胞的体外增殖。
