军团菌在巨噬细胞与中性粒细胞内的生长效应研究

摘    要:目的探索比较军团菌毒性株在巨噬细胞与中性粒细胞内的生长效应.方法诱导分化人的单核细胞系U937细胞使其成为类巨噬细胞,并分离诱导人的外周血单核巨噬细胞以及中性粒细胞;用不同生长时期的军团菌毒力株AA100,感染诱导分化成功的单核巨噬细胞和中性粒细胞,并比较其在巨噬细胞和中性粒细胞中的生长复制状况.结果过对数生长期的军团菌毒力株能够在巨噬细胞内大量复制,扩增状况呈生长阶段的依赖性;而各生长时期的军团菌毒力株均不能在中性粒细胞内大量扩增.结论军团菌逃逸机体的杀伤作用和免疫监视作用与其能够在巨噬细胞内大量复制密切相关.

Abstract:OBJECTIVE To explore and compare the difference of growth kinetics of Legionella virulent strain in the human macrophages and neutrophils. METHODS The human monocyte cell line to macrophage-like cells was induced and the human peripheral monocytes and neutrophils were isolated. The cells were infected by Legionella virulent strain AA100 of different growth phase and its growth and replication status in monocyte-macrophage cells and neutrophils were compared. RESULTS Legionella virulent strain of post-exponential phase could replicate (significantly) in macrophages and its growth was growth-phase dependent.Yet all the growth phases of Legionella could not replicate greatly in neutrophils. CONCLUSIONS The mechanism of Legionella to evade the killing and (immune) surveillance is closely related to its great replication in macrophages.
