固相免疫放射测定法检查乙型肝炎抗原及抗体

摘    要:<正> 目前检出乙型肝炎抗原(HBsAg)及抗体(抗HBs)的新方法较多,主要应用于诊断乙型肝炎;研究乙型肝炎的流行病学;评价药物或疫苗对乙型肝炎的免疫作用,从献血人员或饮食服务人员等健康人群以及胎盘球蛋白等生物制品中,检测乙型肝炎抗原携带情况等。检查乙型肝炎最敏感的方法有放射免疫测定法和反向被动血凝法。我们初步建立了固相免疫放射测定法,并对218份血清及32份胎盘球蛋白进行测定,实践证明此法既简便又灵敏,值得推广应用。本文着重介绍固相免疫放射测定法并与反向被动血凝法做了比较。


Solid-Phase Immunoradioassay For Detecting HBsAg And Anti-HBs
Abstract:A solid-phase immunoradioassay using 125I as the labelling material for hepatitis B antibody was set up in our laboratory. The polyethylene dishs were manufactured by the Third Plastic Factory of Changchun.Method for HBsAg determination and interpretation of the results:1 . First step:Solid-phase antibody + the serum to be determined37℃/over night→solid-phase antibody-antigen complex.2 . Second step:Solid-pha.se antibody-antigen complex + 125I-antibody37℃3hours/or 45℃1.5hours→solid-phase antibody-antigen complex-125I-antibody.Interpretation of the results:The cpm of the serum with a value M + 5·SD of the negative control is considered as positive.Method for anti-HBs determination and interpretation of the results:1 . First step:Solid-phase antibody + the serum to be determined37℃over night→solid-phase antibody-antigen complex.2 . Second step:Solid-phase antibody-antigen complex + serum4℃over night→solid- phase antibody-antigen complex-serum. 3 . Third step:Solid-phase antibody-antigen-serum + 12SI labelled antibodythe number of counting recorded.Interpretation of the results:The numbers of the cpm 40% lower than the control for the serum is considered to be positive.A 36% positive result for HBsAg from 218 "healthy" human sera was obtained,while the positive result for counter passive hemagglutination was only 15%.The solid-phase immunoradioassay method was also used to detect HBsAg in 32 samples of placenta fetal globulin and it was found to be 9.4% positive, while it was negative with counter passive hemagglutination method. Consequently the solid-phase immunoradioassay method is much more sensitive than the counter passive hemagglutination method.
